Using Volatile Buffers

Comparison of the Macherey-Nagel® DEAE-60 to Hydrocell® NS 1500 columns for Phosphorothioate Oligonucleotide [ps-(pT)11T] separations.

Volatile buffers save time for desalting fractions for other studies.

Conditions:

Buffer A: 25 mM triethylamine bicarbonate (~0.35% v/v), 20% MeCN, pH 6.4.
Buffer B: 1M triethylamine bicarbonate (~14% v/v), 20% MeCN, pH 7.6.
Gradient: 1 mL/min., 1%/min., 0 - 100% B.

Sample:

Crude 12mer Thymidine Phosphorothioate Oligonucleotide [ps-(pT)11T].

Note: Failure sequences are not all baseline resolved. The shoulder (35.73 min.) is possibly an isomer of ps-(Tp)11T, since it has the same charge as the 37.1 min.peak and MALDI identification has given conflicting results.
